Abstract. The purpose of this study was to analyze microsatellite loci to assess the genetic polymorphism of Medicago × varia varieties and lines in the breeding process in the Republic of Bashkortostan. Methods. To assess the size and number of SSR loci, 21 samples (lines and varieties) from the Bashkir Research Institute of Agriculture were used. DNA was isolated using the standard CTAB method from 5-7 day-old Medicago × varia sprouts, then SSR-PCR was performed, and the amplification products were separated by electrophoresis in 10 % polyacrylamide gel. Scientific novelty. Alfalfa is one of the most valuable and widespread forage crops used as green fodder, hay, grass meal, haylage, etc., due to its rich content of protein and essential amino acids. In the Republic of Bashkortostan, the most common alfalfa is Medicago sativa L. nothosubsp. varia (Martyn) Arcang, which occurred as a result of crossing several species of alfalfa. Within the framework of this work, for the first time, studies were conducted on the effectiveness of a wide range of microsatellite markers of Medicago sativa for assessing the genetic polymorphism of domestic breeding samples of Medicago × varia. Based on the results of the study, we selected the most informative and effective SSR markers: MsEST53, MSE265, MSE276, MSE470, MSE562 and MSE590, which can be used to assess genetic polymorphism and further certification of Medicago × varia varieties and lines.
