Abstract. The purpose of the study was to conduct a comprehensive assessment of the genetic safety and functional reliability of recombinant probiotic strains in veterinary practice using a biocorrective agent based on the recombinant strain Lactobacillus plantarum B-11007 carrying the plasmid construct pLacDualIL-10/IL-22 for the expression of immunomodulatory cytokines IL-10 and IL-22. Genetic safety was considered in terms of plasmid stability and the risk of horizontal gene transfer, while functional reliability was evaluated through the reproducibility of the immunomodulatory effect. Methods. The study was conducted in 2024–2026 at the Genome Center of the North-Caucasus Federal University and the laboratory of the Stavropol Agrarian University. The study assessed the probability of horizontal transfer of plasmid DNA to other bacteria, taking into account phylogenetic distance and the absence of conjugative and mobilization genes. Simulated gastric and intestinal conditions were performed using pepsin, pancreatin, and bile acids to evaluate DNA degradation. Additionally, local immunomodulatory effects of the synthesized cytokines on the intestinal mucosa of calves were investigated. Results. The strain demonstrated high structural and functional stability, a minimal risk of intergeneric plasmid DNA transfer, and pronounced local immunomodulatory activity. Strengthening of the epithelial barrier function, stimulation of antimicrobial peptide synthesis, and anti-inflammatory effects were observed, with no systemic circulation or accumulation of cytokines in the body. Scientific novelty. This study provides, for the first time, a comprehensive evaluation of the genetic biosafety and functional reliability of the recombinant probiotic strain Lactobacillus plantarum B-11007/pLacDualIL-10/IL-22, considering horizontal gene transfer risk, plasmid stability, and localized immunomodulatory activity. Conclusion. The obtained results confirm the safety and efficacy of the recombinant strain for use in calves and establish a scientific basis for the development of next-generation biocorrective probiotic agents aimed at targeted immunomodulation and enhancement of intestinal microbiota stability.
